    Hi,

    I am starting this new thread as I didn't find much information regarding PGD.I am 33 years old, married for 6 years.Trying to conceive for almost 4 years. All test of myself and Husband are normal. Did 5 IUI's and 5 IVF's (2 fresh and 3 frozen) in Pune.2 transfer are Day 3 and 3 Day 5 . After facing so many failures trying to build my hope again. This time we started seeing Dr.Firuza Parikh in Jaslok Hospital. She suggested I do another IVF cycle with ERA and PGS. Just curious to know if anyone has done this before. It would be really helpful if you share your thoughts regarding this.

    Hi @yogita04

    Iam from Mumbai. I had met firuza last year.
    Now regarding ur query , while I was consulting her , it's a huge rush and the wait is long , so I made a friend there who came all the way from chennai for IVF to her. She was in her 3rd IVF with frozen. The cycle failed and then she was advised for the ERA. This test is basically to know why the embryos aren't implanting into the endometrium. They give u low dose stims in this procedure.
    Pgs is not done everywhere in Mumbai . She does it .. This is identify genetically abnormal embryos which may appear of top quality under the microscope.

    Hello @yogita04

    I had ERA. Its used to identity the receptiveness of endometrium . It gives an idea whether the endometrium is pre receptive, receptive or post receptive. With the help of this test Dr's try to time the transfer the embryo when your lining to receptive to embryo. For this test Dr will give tablets to thicken the lining so from n no of days and after that introduce progestrone supplements and around day 15-18( not exactly sure) they will take a biopsy of the lining. If your cervix behaves well Biopsy procedure is not painful at all.
    In PGS/PGD the embroyologist will remove a single cell from the embryo and will check for any chromosomal abnormality or genetic disease(inherited) respy. I remember reading PGS gives a better result when done on day 5 embryo. The main draw back is you need a decent no of embryos(>4) to go through this process as sometimes removing a cell from embryo can affect its quality.
    I hope you have ruled out Karotype problems, lining issues, clotting and immune problems before considering Psg.

    Good Luck!!

    Thank you @Agony and @Maggilg for your reply.

    Magglig yes we have done Karyotyping for myself and Husband.. Its normal too.. Also my reports of Antibodies are normal. Our's is unexplained infertility so dnt know how much PGS is going to help. But we have decided to move further with the test. It is very expensive test so still having doubts.Having my pickup scheduled on Wednesday..Hope this cycle works for me..Leaving everything on Gods hand now..

    Baby dust to all beautiful ladies who are going through this difficult journey.

    @yogita04 I had done ERA too. You can say it's a mock ET cycle, where all medications that one goes through during an ET is done. On the day when ET is supposed to be done, they do a biopsy to retrieve endometrium tissues. This helps identify implantation window - whether endometrium is receptive pre-receptive, post-receptive, non-receptive etc. It's result will help doctors time your ET. Its a costly affair and suggested to ppl with 3+ ivf failures.

    All the best for your further treatments. Let us know your experience with Dr. Firoza Shaikh

    @yogita04,
    Sometimes even with normal karyotype the eggs or sperm produced or the embryo might be abnormal. It's quite normal for a embryo to be abnormal (reason for miscarriage before 5 weeks,I.e even before knowing we r pregnant) and it's slightly more in Ivf that's why even after producing 7-10 eggs we end up getting 2-4 good embryos on day 5. Pgs helps us to determine whether the embryos produced are chromosomal euploid. So I think that's why dr has recommended it.
    Did u have a hysteroscopy to check uterine lining ?? Do u get recommended thickness and triple lining before transfer ? You mentioned all ur antibodies are normal do those include thromboplia profile (clotting and sticky blood) and uterine killer cells.? Sorry to b specific. If u r not knee to try Pgs u can try embryoscope (or can use embryos cope along with Pgs) it provides a std envy for embryos to grow as there is no need to take them out for growth inspection and embryologist can monitor the progress as growth of embryo is video recorded. Also try not to push ur self and give a break of at least 3 cycles before you go for another cycle after a failed one. And one more main culprit in ttc in Stress. It does more harm than u think. Try to focus ur mind into positive things and keep the stress bit*h at bay.
    Sometimes Even though everything is normal a cycle may not work out. We just have to keep trying. I hope this cycle brings baby dust to you.
